When Harry changed into Goyle (Crabbe?) he didn't need his glasses anymore because the DNA strand (or chromosome, depending on how you want to look at it) no longer had whatever problem caused his weak eyesight. I'd assume that it would change whatever DNA strand or chromosome the lycanthropy is attached to.

I think the question is the other way around. Not if a person drinks polyjuice potion containing werewolf hair, but a werewolf drinking polyjuice potion.

In the movie, Hermione read out of the book (I don't remember if that happened in the book) that the potion will only last for one hour. I don't think it matters if the wizard is more experianced or not. Moody kept a hip flask on him at all times, so Barty Jr. used that to his advantage. He just kept the flask filled up with Polyjuice.
